제목 Intervening Effects of Gut Microbiota and Metabolites on Obesity-Related Glomerulopathy Based on Metabolomics, 16S rRNA Sequencing, and Metorigin Analyses

초록 Objectives: As a secondary glomerular disease, Obesity-related glomerulopathy (ORG) can occur in people with body mass index ≥30kg/m². Abnormal lipid metabolism, disturbance of intestinal flora and inflammation critically contribute to the pathogenesis of ORG. The deteriorating process of ORG, in which a complex network of metabolites and microorganism events was involved, is not fully elucidated yet. In this study, we aimed to reveal the role of alter metabolites and intestinal microbiota in ORG. Methods: Six-week-old C57BL/6J mice were used to establish ORG model via high-fat diet (HFD) feeding and the pathological injury of kidney were assessed by biochemical-specific markers and histological staining assays. Here we employed liquid chromatography with tandem mass spectrometry (LC-MS/MS)-based metabolomic, 16S rRNA Sequencing and Metorigin analyst to measure the changes of metabolites expression and intestinal flora in ORG model mice. Results: The HFD-fed mice showed obvious proteinuria and glomerulomegaly as compared to normal diet (ND) mice. According to our results, we found that leukotriene C4 were increased in the HFD group, while alpha-linolenic acid (ALA), D-Glutamine, Linoleic acid (LA) were decreased. Furthermore, metabolic reprogramming was occurred in ORG mice, including glycerophospholipid and Linoleic acid metabolism, which are closely related to the disturbulance of Parabacteroides goldsteinii. Conclusions: Our study identified the decrease of Parabacteroides goldsteinii interferes with glycerophospholipid and linoleic acid metabolism, which contribute to lipid accumulation and inflammation. The present study provides a new potential intervention target for the treatment of pancreatic cancer in clinical practice.
