| 초록 | Background: IL-18BP binds to IL-18 and neutralizes its biological activity. This protein is constitutively expressed in mononuclear cells and can be enhanced by IFN-gamma. However, little is known about which cytokines and cellular types are responsible for IL-18BP secretion in kidney cells. The aim of this study was to examine whether IL-12, IL-17, IL-27 and interferon gamma induce IL-18BPa secretion in HK-2 cells and human podocytes. Methods: We stimulated HK-2 cells and human podocytes with IL-12, IL-17, IL-27 and interferon gamma to examine IL-18BP secretion. Using qRT-PCR and ELISA, mRNA expression and secretion of IL18BPa from these cell lines were measured. Results: Of the investigated cytokines, only interferon gamma and IL-27 increased mRNA expression of IL-18BPa. Since IL-18BP is a secretory protein, its level in supernatants of HK-2 cells and human podocytes were examined under by ELISA. IL-12 and IL-17, previously known as IL-18BP inducing cytokines, did not stimulate its secretion in both HK-2 and human podocytes, but interferon gamma and IL-27 enhanced release of IL-18BP from both HK-2 and human podocytes in time and dose-dependent manner. Conclusion: Our results demonstrate for the first time that IL-18BPa is expressed in HK-2 cells and human podocytes. Interleukin-27 is a new cytokine that induces expression of IL-18BPa in HK-2 cells and human podocytes. |
